چکیده

The EIF4E gene spans >50 kbp and contains 8 exons, one of them being alternative. It codes for the major, 4749 nucleotides long transcript variant 1, that codes 217 aa long protein. The longest transcript, variant 2 (4842 nucleotides) contains an additional in-frame exon in 3' coding region compared to variant 1 and codes for a protein (248 aa long, isoform 2) with a longer cterminus compared to isoform 1. Transcript variant 3 is 3406 nucleotide long and uses an alternative exon for the 5'UTR and 5' coding region that results in translation initiation from a distinct ATG, and an isoform 3, (237 aa) with a longer and distinct Nterminus compared to isoform 1. Transcript variants 2 and 3 are predicted from cDNA sequences, but their expression at mRNA or protein levels was not studied.

Multiple mRNAs encode the murine translation initiation factor eIF-4E.

All eukaryotic cellular mRNAs (except organellar) possess at their 5' end the structure m7GpppX (where X is any nucleotide) termed the "cap." The cap structure facilitates the melting of mRNA 5' secondary structure through the action of initiation factor-4F (eIF-4F) in conjunction with eIF-4B. eIF-4F consists of three subunits of which one, eIF-4E (eIF-4E has recently been designated eIF-4 alph...

The translation initiation factor eIF-4E binds to a common motif shared by the translation factor eIF-4 gamma and the translational repressors 4E-binding proteins.

Eukaryotic translation initiation factor 4E (eIF-4E), which possesses cap-binding activity, functions in the recruitment of mRNA to polysomes as part of a three-subunit complex, eIF-4F (cap-binding complex). eIF-4E is the least abundant of all translation initiation factors and a target of growth regulatory pathways. Recently, two human cDNAs encoding novel eIF-4E-binding proteins (4E-BPs) whic...

Cell cycle progression and proliferation despite 4BP-1 dephosphorylation.

Proliferation and cell cycle progression in response to growth factors require de novo protein synthesis. It has been proposed that binding of the eukaryotic translation initiation factor 4E (eIF-4E) to the inhibitory protein 4BP-1 blocks translation by preventing access of eIF-4G to the 5' cap of the mRNA. The signal for translation initiation is thought to involve phosphorylation of 4BP-1, wh...

ATP-dependent unwinding of messenger RNA structure by eukaryotic initiation factors.

Interaction of protein synthesis initiation factors with mRNA has been studied in order to characterize early events in the eukaryotic translation pathway. Individual reovirus mRNAs labeled with 32P in the alpha position relative to the m7G cap and eukaryotic initiation factor (eIF)-4A, -4B, and -4F purified from rabbit reticulocytes were employed. It was found that eIF-4A causes a structural c...

Cloning of eukaryotic protein synthesis initiation factor genes: isolation and characterization of cDNA clones encoding factor eIF-4A.

Monoclonal antibodies directed against rabbit reticulocyte protein synthesis initiation factor 4A (eIF-4A) were used to isolate mouse cDNA clones expressing eIF-4A protein sequences in E. coli. The identity of cDNA clones encoding eIF-4A sequences was confirmed by hybrid-selected translation and peptide mapping of the translation product. Analysis of the mRNA coding for eIF-4A from mouse liver ...
